联合应用柚皮苷及黄芩苷对人牙周膜细胞生物学活性的影响

Effects of naringin and baicalin combination on the biological activity of human periodontal ligament cells

目的:观察中药有效成分柚皮苷和黄芩苷联合应用对人牙周膜细胞(hPDLCs)生物学活性的作用。方法:用酶结合组织块法原代培养hPDLCs;通过MTT法、酶联免疫吸附法、诱导矿化实验测定二者配比之后对hPDLCs增殖、Ⅰ型胶原蛋白的表达、碱性磷酸酶(ALP)活性及钙化结节形成的影响。结果:1.0 mg/L的柚皮苷与0.1 mg/L黄芩苷配伍对细胞增殖、Ⅰ型胶原蛋白表达、ALP活性以及矿化的促进作用明显。结论:柚皮苷与黄芩苷配伍组合可增强hPDLCs的成骨能力。

Objective：To study the effects of naringin and baicalin combination on the biological activity of human periodontal ligament cells（hPDLCs）.Methods：hPDLCs were cultured and identified in vitro.The cells were exposed to naringin,baicalin or combination of the both at various concentrations.The proliferation,collagen-Ⅰ protein expression,alkaline phosphatase（ALP）activity and mineralization of hPDLCs were examined by MTT assay,enzyme linked immunosorbent assay and Von Kossa staining.Results：Naringin at 1.0 mg/L combined with baicalin at 0.1 mg/L showed the strongest promotion effects on the proliferation,collagen-Ⅰ protein expression,ALP activity and mineralization of hPDLCs.Conclusion：Naringin and baicalin combination may play a synergistic effect in the bone formation of hPDLCs.
