摘要
目的:探讨Fascin-1和MMP-1在喉鳞状细胞癌(LSCC)和癌旁组织中的表达及意义。方法:免疫组化SP法检测50例喉鳞状细胞癌及其癌旁0.5cm、1.0cm、2.0cm组织中Fascin-1和MMP-1的表达情况,并分析两者的关系。结果:Fascin-1在喉鳞癌中的表达及癌旁0.5cm、1.0cm及2.0cm的黏膜中表达率依次降低,分别为80.0%、27.5%、22.5%、4.0%,除了癌旁0.5组与癌旁1.0组,癌旁1.0组与癌旁2.0组差别无统计学意义之外(P>0.008),其余各组间差别均有统计学意义(P<0.008)。MMP-1在喉癌癌旁0.5cm、1.0cm及2.0cm的表达率依次降低,分别为85.0%、35.0%、17.5%、5.0%,除了癌旁0.5组与癌旁1.0组,癌旁1.0与癌旁2.0差别无统计学意义之外(P>0.008),其余各组间差别均有统计学意义(P<0.008)。喉鳞癌中Fascin-1蛋白的表达与癌的外科切缘,临床分期,肿瘤分化程度及淋巴结转移相关,与性别和年龄无关。MMP-1蛋白表达同样与癌的外科切缘,临床分期,肿瘤的分化程度及淋巴结转移相关,与性别和年龄无关。二者的表达呈正相关(P<0.001)。结论:Fascin-1和MMP-1在喉鳞癌侵袭和转移中发挥重要作用,二者联合检测可能成为预测喉鳞癌预后的生物学指标,并为以Fascin-1和MMP-1为靶点的癌症治疗提供了思路。
Objective:To study the expression of Fascin-1 and MMP-1(matrix metalloproteinase-1) in laryngeal squamous cell carcinoma(LSCC) and adjacent tissues and their clinical significance.Methods: Using citric-SP-immunochemical technique,the expression of Fascin-1 and MMP-1 were studied in different regions of larynx squamous cell carcinoma including 40 cases laryngeal carcinoma and adjacent tissues which was 0.5,1.0,2.0 cm away from cancer tissue.Results: The expression of Fascin-1 reduced in sequence cancer tissue,at 0.5 cm,1.0 cm,2.0cm adjacent to carcinoma,which were 80.0%,27.5%,22.5%,4.0%,there was no significant difference in rate of addition(P0.008) in group of 1.0cm and 2.0 cm and the group of 0.5cm and 1.0cm,the other groups differences were statistically significant(P0.008).The expression rates of MMP-1 in cancer tissue,at 0.5cm,1.0cm,2.0cm adjacent tissaes were 85.0%,35.0%,17.5%,5.0%,there was no significant difference(P0.008) group between 1.0cm and 2.0 cm and the group of 0.5cm and 1.0cm,in other groups differences were statistically significant(P0.008).The expression of Fascin-1 protein in Laryngeal squamous cell carcinoma had a relationship with clinical stage of cancer,tumor location,differentiation and lymph node metastasis,but not associated with age and gender.The expression of MMP-1 have a relationship with clinical stage of cancer,tumor location,tumor differentiation and lymph node metastasis,but not associated with age and gender.the expression of Fascin-1 and MMP-1 were positively correlated(P0.001).Conclusion:Fascin-1 and MMP-1 maybe directly associated with the development of LSCC,which may play an important role in the process of invasion and metastasis of LSCC,combined detection of Fascin-1 and MMP-1 may be an important biological factor affecting the prognosis judgement of LSCC.
