摘要
目的:研究宫颈腺癌1p31杂合性丢失的发生率,探讨在此位点的ARHI基因在宫颈腺癌发生发展中的作用。方法:采用PCR银染技术检测30例宫颈腺癌中1p31杂合性丢失的发生率,其中Ⅰ,Ⅱ期18例,Ⅲ、Ⅳ期12例,免疫组化方法检测在此位点的ARHI蛋白的表达。结果:30例宫颈腺癌组织中LOH阳性的比例为63.33%(19/30),Ⅲ期、Ⅳ期LOH阳性率明显高于Ⅰ,Ⅱ期,差异有统计学意义(χ2=5.98,P<0.05)。在高、中、低分化癌中无统计学差异(χ2=4.88,P>0.05),ARHI蛋白的阳性表达率与淋巴结转移无关,差异无统计学意义(χ2=2.16,P>0.05)。宫颈腺癌组织中ARHI表达(46.67%,14/30)低于正常宫颈组织(90.0%,27/30),差异具有统计学意义(χ2=13.02,P<0.05)。结论:1p31位点可能存在宫颈腺癌相关抑癌基因,ARHI基因在宫颈腺癌发生发展过程中起重要作用。
Objective:To study the incidence of loss of 1p31 heterozygosity and explore the effect of ARHI gene in the locus on the occurrence and development of cervical adenocarcinoma.Methods:PCR silver staining technique was used to detect the incidence of loss of 1p31 heterozygosity in 30 cases with cervical adenocarcinoma,including 18 cases at stage Ⅰand stage Ⅱ,12 cases at stage Ⅲ and stage Ⅳ.Immunohistochemical method was used to detect the expression of ARHI protein in the locus.Results:The proportion of loss of heterozygosity in 30 cases with cervical adenocarcinoma was 63.33%(19/30),the positive rates of loss of heterozygosity at stage Ⅲ and stage Ⅳ were significantly higher than those at stage Ⅰand stage Ⅱ(χ2=5.98,P0.05).There was no significant difference in the positive rate of loss of heterozygosity among poorly differentiated cervical adenocarcinoma,moderately differentiated cervical adenocarcinoma and well differentiated cervical adenocarcinoma(χ2=4.88,P0.05).There was no correlation between the positive expression rate of ARHI protein and lymph node metastasis(χ2=2.16,P0.05).The expression rate of ARHI protein in cervical adenocarcinoma tissue was 46.67%(14/30),which was significantly higher than that in normal cervical tissue(90.0%,27/30)(χ2=13.02,P0.05).Conclusion:Cervical adenocarcinoma related tumor suppressor gene may exist in 1p31 locus,RHI gene plays an important role in the occurrence and development of cervical adenocarcinoma.
