T-bet/GATA-3在子痫前期患者蜕膜组织中的表达

Expression of T-bet/GATA-3 in decidual tissue of patients with preeclampsia

目的:比较T-bet/CATA-3在正常妊娠和子痫前期患者蜕膜组织中的表达水平,并探讨两者与子痫前期发病的关系。方法:采用病例对照研究方法,选择轻度子痫前期和重度子痫前期患者各13例作为研究组,选择14例正常妊娠孕妇为对照组,用Western bloting方法比较三组孕妇蜕膜中T-bet/GATA-3的表达水平。结果:①研究组T-bet表达水平比对照组高,重度子痫前期组比轻度组高,各组比较差异有统计学意义(P<0.05)。②研究组的GATA-3表达比对照组降低,重度子痫前期组比轻度组降低。各组比较差异均有统计学意义。③三组T-bet/GATA-3的比值比较差异有统计学意义,且随着病情的加重T-bet/CATA-3比值增大。④T-bet与GATA-3存在明显的负相关性(r=-0.895,P<0.01)。结论:T-bet和GATA-3的表达失衡和紊乱可能是子痫前期免疫学发病机制中的重要环节,Th1/Th2的失衡可能是妊娠期高血压疾病发生机制之一。

Objective：To compare the expression levels of T-bet/GATA-3 in decidual tissues between normal pregnant women and patients with preeclampsia,explore their relationships with the occurrence of preeclampsia. Methods：A case-control study was performed,13 patients with mild preeclampsia and 13 patients with severe preeclampsia were selected as study group,14 normal pregnant women were selected as control group.Western blotting was used to compare the expression levels of T-bet/GATA-3 in the three groups. Results：The expression level of T-bet in study group was higher than that in control group,the expression level of T-bet in severe preeclampsia group was higher than that in mild preeclampsia group,there was significant difference（P0.05）.The expression level of GATA-3 in study group was lower than that in control group,the expression level of GATA-3 in severe preeclampsia group was lower than that in mild preeclampsia group,there was significant difference.There was significant difference in the ratio of T-bet/GATA-3 among the three groups,and the ratio of T-bet/GATA-3 increased with the aggravation of the disease.There was a significant negative correlation between T-bet and GATA-3（γ=-0.895,P0.05）. Conclusion：The unbalance and disorder of T-bet expression and GATA-3 expression is an important link in the immune pathogenesis of preeclampsia,Thl/Th2 unbalance may be a pathogenesis of hypertensive disorder complicating pregnancy.
