摘要
目的通过对2种方法进行比较,探讨糖尿病动物模型的建立方法。方法用四氧嘧啶和链佐霉素分别对小鼠进行腹腔注射,建立糖尿病动物模型,链佐霉素组造模前小鼠高脂高糖喂养。小鼠分两组:一组单独用四氧嘧啶腹腔注射一次;二组用链佐霉素单独腹腔注射,小剂量多次;每组还设一个正常组作对照,测定造模前、后血糖含量及结束时血清甘油三酯(TG)、总胆固醇(TC)含量。结果一组、二组造模前、后血糖含量依次为:(5.95±0.48)、(13.38±4.35);(6.06±0.60)、(12.98±3.34)(;mmol/L),两组前后比较均为P<0.01,差异明显;正常对照组前后无明显差异;一组、二组(每组包括正常组和模型组)总胆固醇(TC)分别为(2.81±0.40)、(2.60±0.47);(2.29±0.39)、(2.87±0.53()mmol/L)(二组前后比较P<0.05,差异明显);一、二组造模成功率分别为51.4%、60.6%;死亡率分别为14.3%和5.7%;血清甘油三酯两组无明显差异。结论两种方法造模均成功,造模后血糖均明显升高,但使用链佐霉素造模成功率高于使用四氧嘧啶,且总胆固醇升高较明显,死亡率较低。
Objective Through 2 kinds of methods are compared,the diabetes mellitus animal model.Methods With alloxan and streptozotocin on mice were intraperitoneally,establishment of animal model of diabetes mellitus,streptozotocin group before making model mice high-fat feeding.Mice were divided into two groups:a group of separate with alloxan injected intraperitoneally one;two groups with single intraperitoneal injection of streptozotocin,multiple low dose;each group is also provided with a normal control groups.Determination before making model,after the end of the content of blood sugar and serum triglyceride (TG),total cholesterol (TC) content.Results Of a group,group two before making model,sugar content as follows:(5.95±0.48),(13.38±4.35);(6.06±0.60),(12.98±3.34);(mmol/L),the two groups before and after comparison of all P 0.01,difference is apparent;the normal control group and no significant difference;a group,two groups of (each including normal group and model group) and total cholesterol (TC) were(2.81±0.40),(2.60±0.47);(2.29±0.39),(2.87±0.53)(mmol/L) (P0.05 in two groups before and after comparison,difference is apparent);one or two groups of modeling success rates were 51.4%,60.6% respectively;mortality rate 14.3% and 5.7%;serum triglyceride had no difference between the two groups;Conclusion Two methods of molding were successful,after building the model of blood glucose were significantly increased,but the use of streptozotocin modeling success rate higher than the use of alloxan,and total cholesterol increased significantly,lower mortality.
