摘要
目的研究紫心甘薯总黄酮(PSPF)的降糖及其抗氧化作用。方法采用链脲佐菌素(STZ,65 mg.kg-1)腹腔注射诱导形成糖尿病大鼠模型,实验组大鼠分为正常对照组、糖尿病模型组、PSPF高剂量组(240 mg.kg-1),中剂量组(120 mg.kg-1),低剂量组(60 mg.kg-1),阳性对照组(二甲双胍,90 mg.kg-1),PSPF灌胃4周,每天测量体重、饮食和饮水量,初期、中期、后期测量空腹血糖(FBG),最后取血测定糖化血清蛋白(GSP)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、超氧化物歧化酶(T-SOD)的变化并测量餐后2 h血糖(2 h BG)。结果 PSPF灌胃4周后,糖尿病大鼠"三多一少"症状明显改善,与模型组相比,高、中灌胃组均有效降低FBG、GSP、2 h BG、MDA,提高大鼠血清中的T-SOD、GSH含量(P<0.01);低剂量灌胃组均可明显改善FBG、GSP(P<0.05)、MDA、T-SOD含量(P<0.01);血糖与抗氧化指标统计学分析相关性显著(P<0.05)。结论 PSPF可以明显改善糖尿病大鼠的血糖异常,提高大鼠抗氧化能力。
OBJECTIVE To study hypoglycemic and antioxidant effects of purple sweet potato flavonoids(PSPF).METHODS Diabetes mellitus(DM) was induced in rats by intraperitoneal injection of streptozotocin(STZ,65 mg·kg-1).All the rats were divided into six groups randomly,including normal control group,diabetic model group,metformin group(90 mg·kg-1),PSPF high-dose(240 mg·kg-1),medium dose(120 mg·kg-1) and low dose group(60 mg·kg-1),weight,food and fluid intake were monitored every day,fasting blood glucose(FBG)was measured every 2 week and at the end of the experiment,GSP,MDA,GSH,T-SOD changes were tested.Also,blood glucose 2 h after meal(2 h BG)was measured.RESULTS Compared with diabetic model group,diabetic symptoms were ameliorated after rats were fed with PSPF.The levels of blood glucose,GSP,2 h BG,MDA were decreased and serum T-SOD,GSH were increased remarkably in the groups of PSPF 120 and 240 mg·kg-1(P0.01);the levels of blood glucose,serum GSP(P0.05),MDA,T-SOD(P0.01) were improved in the group of PSPF 60 mg·kg-1.CONCLUSION Purple sweet potato flavonoids can decrease the blood glucose level and increase antioxidant capacity of diabetic rats.
