血府逐瘀汤诱导内皮细胞血管新生中一氧化氮的作用 被引量:34

Effect of Xuefu Zhuyu Decoction on the Nitric oxide in the Endothelial Angiogenesis

摘要 目的研究血府逐瘀汤促进内皮细胞参与血管新生的作用机制。方法选择SD大鼠12只,随机分为血府逐瘀汤组和空白对照血,每组6只。分别制备血府逐瘀汤含药血清和空白对照血清,采用1.25%、2.5%、5.0%不同浓度血府逐瘀汤含药血清和空白对照血清诱导内皮细胞48h,检测不同浓度血清对内皮细胞ECV304增殖、迁移和体外成血管的影响,并检测细胞培养上清液中一氧化氮(NO)浓度、胞内NO浓度和内皮型一氧化氮合成酶(eNOS)活性及表达。结果与空白对照组相同血清含量比较,1.25%、2.5%药物浓度下细胞增殖明显增加(P<0.05),2.5%、5.0%药物浓度下迁移细胞数明显增加(P<0.01),各药物浓度下管腔个数均明显增加(P<0.01);各浓度药物均可显著提高内皮来源的胞内、胞外NO浓度及eNOS表达(P<0.05或P<0.01),1.25%、2.5%浓度药物还可显著提高eNOS的活性(P<0.05或P<0.01)。结论血府逐瘀汤通过促进eNOS的表达和活性,提高胞内、外气体分子NO水平发挥促血管新生作用。 Objective To investigate the mechanism of chest) in promoting the endothelia-derived angiogenesis. Xuefu Zhuyu Decoction (Decoction for removing blood stasis in the Methods Twelve SD rats were randomized into Xuefu Zhuyu Decoction group and blank control group with 6 in each. The medicine-containing serum with the Xuefu Zhuyu Decoction ingredients in it and the blank control serum were prepared respectively. By using a serum pharmacology technique, the 1.25%, 2.5%, and 5% concentrations of Xuefu Zhuyu Decoction containing serum and the blank control serum were adopted for endothelial cell incubation for 48h. The effect on the reproduction, migration, and in vitro vasoformation of endothelial cells ECV304 were examined. Moreover, the nitric oxide (NO) concentration in the cultured supernate was evaluated, the intraeellular NO concentration and endothelial nitric oxide synthetase (eNOS) activity were studied, and the eNOS expression was determined. Results Compared with the same concentration serum of the blank control group, the reproduction of ceils under the condition of low (1.25%) and medium (2.5%) concentrations of medicine-containing serum was significantly increased (P〈0.05), the migration of cells under the condition of medium (2.5%) and high (5%) concentrations of medicine-containing serum was significantly increased (P〈0.01). The lumens under the condition of all concentrations of medicine-containing serum were significantly increased (P〈0.01). All concentrations of medicinal serum could increase the endothelia-derived intracellular and extracellular NO concentrations and the eNOS expression (P〈0.05 or P〈0.01). And the 1.25% and 2.5 % concentrations of medicine-containing serum could enhance the eNOS activity as well (P〈0.05 or P〈0.01). Conclusion Xuefu Zhuyu Decoction increases the intracellular and extracellular NO concentration of gas to promote angiogenesis through improving the eNOS expression and activity.
