降解纤维素真菌的分离鉴定及其产酶条件的研究

摘要 从腐烂的木头、堆肥及土壤等样品中,通过刚果红染色鉴定和液体发酵培养分离筛选得到2株高产纤维素酶青霉菌株A2和A6,对其产酶条件进行初步优化。结果表明,A2和A6羧甲基纤维素酶活分别为280.14,247.58U;A2最佳产酶条件为接种量15%(V/V),初始pH 5,时间120h,添加一定量Mn2+和Ca2+到培养基中酶活分别升高10.56%和7.95%,加入Fe2+和Hg2+对酶活有一定抑制;A6最佳产酶条件为接种量15%(V/V),初始pH 5,时间96h,添加一定量Mn2+到培养基中酶活升高13.1%,而加入Ca2+、Fe2+和Hg2+对酶活有一定抑制。 Two Penicilliums named A2and A6with high cellulose ac-tivity were screened from the rotten wood,compost and soil by Con-go red staining and Liquid fermentation,the CMCase activity of A2 and A6is 280.14Uand 247.58U,and the optimal fermentation conditions were obtained by experiments.The optimal fermentation conditions of A2were as follows:inoculum number 15%,initial pH 5,culture time 120h,the CMCase activities increasing 11.35% and 7.80%with Mn2+and Ca2+addition,and reducing with the me-dium added Fe2+and Hg2+addtion;The optimal fermentation con-ditions of A6were as follows:inoculum number 15%,initial pH 5,culture time 96h,the CMCase activity increasing 13.92% with Mn2+addition,and reducing with Ca2+,Fe2+and Hg2+addition.
