大豆异黄酮联合顺铂对A549细胞增殖和凋亡的影响

EFFECT OF SOYBEAN ISOFLAVONES COMBINED WITH CISPLATIN ON PROLIFERATION AND APOPTOSIS OF A549 CELL

目的探讨大豆异黄酮(ISF)联合顺铂(DDP)化疗对A549肺癌细胞增殖和凋亡的影响。方法采用噻唑蓝(MTT)法观察ISF和DDP联用对A549肺癌细胞增殖的影响,并计算联合指数(Q值)判定联合用药的相互作用,同时采用TUNEL染色法检测二者对A549细胞凋亡的影响。结果 ISF和DDP单独使用均可时间和浓度依赖性地抑制肺癌A549细胞的增殖。DDP浓度在3 mg/L时,ISF和DDP联用后抗细胞增殖作用增强,二者有相加或协同作用,且在一定浓度范围内,联合用药对细胞生长的抑制作用呈量效关系。ISF单用或和DDP联用均可浓度依赖性地诱导肺癌细胞凋亡。结论 ISF可通过诱导A549细胞凋亡而发挥抗肺癌作用;ISF和DDP联用可增强对肺癌A549细胞增殖的抑制和凋亡的诱导,二者具有协同抗肿瘤作用。

Objective To investigate the inhibitory effect and mechanism of soybean isoflavones（ISF） combined with cisplatin（DDP） on proliferation and apoptosis of A549 lung carcinoma cells.Method Methylthiazolyl tetrazolium（MTT） assay was used to examine the effects of ISF and DDP on proliferation of A549 cells,and the multidrug effect was analyzed by calculating Q value.The terminal deoxynucleotidyl transferase mediated nick end labeling（TUNEL） method was used for determination of A549 cell apoptosis.Result ISF and DDP inhibited the proliferation of lung carcinoma cells in a time-and concentration-dependent manner.The synergistic or additive effect on inhibition of A549 proliferation was detected in combination with ISF and DDP when the concentration of DDP was 3 mg/L;and the concentration-dependent pattern was detected within certain concentration of ISF.Furthermore,single ISF or ISF combined with DDP induced apoptosis of A549 lung carcinoma cell in concentration-dependent manner.Conclusion ISF could exert anti-lung cancer effect via induction of cell apoptosis.ISF combined with DDP can play synergistic anti-tumor effect by enhancing growth inhibition and apoptosis on implanted A549 lung carcinoma cell.
