延衰合剂对D-半乳糖诱导的亚急性衰老小鼠免疫功能影响的实验研究 被引量:2

Effect of Yanshuai Mixture on Immune Function in Subacute Aging Mice Caused by D-galactose

摘要 目的研究延衰合剂(yanshuai mixture,YSM)对D-半乳糖所致亚急性衰老小鼠胸腺、脾脏组织结构及白介素2水平的影响。方法选昆明系小鼠,用D-半乳糖建立衰老模型。应用光镜、电镜技术观察延衰合剂治疗前后衰老小鼠胸腺、脾脏的形态学改变;检测胸腺指数及脾脏指数;酶联免疫分析法检测血清白介素2的变化。结果衰老模型组小鼠胸腺细胞排列紊乱,线粒体肿胀,脾脏淋巴细胞形态异常,胸腺脾脏指数降低,血清白介素2水平明显降低(模型组与正常对照组比较:p<0.05);用药后,胸腺细胞形态结构改善,排列趋于正常,脾淋巴细胞形态接近规则,血清白介素2水平增加(p<0.05)。延衰合剂高剂量组作用优于补肾益寿胶囊组及延衰合剂低中剂量组(p<0.05)。结论延衰合剂可以改善衰老小鼠胸腺、脾脏的形态结构,抑制衰老小鼠血清白介素2水平的下降,具有一定的延缓衰老的作用。 Objective To study the effect of yanshuai mixture on the morphology of thymus,spleen and serum interleukin 2 in subacute aging mice. Methods The Kunming mice were selected to build aging mice model by D-galactose. The changes in morphology of thymus and spleen were observed by light microscope and electron microscopy after treatment with yanshual mixture. The indexes of thymus and spleen were measured,The changes of serum interleukin 2 were determined by ELISA. Results The thymus cells arranged disorderly, mitochondria swelled, and the splenic lymphocytes have abnormal morphology, the indexes of thymus and spleen were reduced, the serum interleukin 2 level decreased in aging mice, having significant difference between model group and normal control group ( P 〈 0. 05). After treatment, the morphology structure of thymus cells improved, histological structure of the splenic lymphocytes improved and the serum intedeukin 2 increased ( P 〈 0. 05). The effect of yanshuai mixture high-dosage group were better than that of Bushenyishou Capsules group, yanshuai mixture lowdosage group and yanshuai mixture middle-dosage group( P 〈 0. 05 ). Conclusion Yanshuai mixture could improve the morphology of thymus and spleen in aging mice and could inhibit the decreasing of serum interleukin 2 of aging mice,resuh in the delay of aging in mice.
