摘要
目的探讨5-氟尿嘧啶磁性白蛋白微球(5-Fu—MAMS)联合恒定外磁场体外对人胰腺癌PC-3细胞生长的抑制作用及其机制。方法将体外培养人胰腺癌PC-3细胞分为5-Fu—MAMS联合磁场组;5-Fu-MAMS组;游离5-Fu组,倒置显微镜下观察肿瘤细胞的生长状态,噻唑蓝(MTT)比色分析法检测各组细胞的抑制率。结果5-Fu浓度为20.0mg/L时,5-Fu—MAMS组与游离5-Fu组抑制率分别为57.7%和59.6%,IR〉50%,药物敏感,差异无统计学意义(P〉0.05);5-Fu浓度为10.0mg/L时,5-Fu-MAMS联合磁场能明显提高化疗药物的抑瘤效果,IR为60%,IR与其他两组比较差异有统计学意义(P〈0.05)。结论5-Fu—MAMS和游离5-Fu有相似的药理作用;联合恒定外磁场能显著增强5-Fu—MAMS的抑瘤效应。
Objective To investigate the growth inhibiting action of 5-fluorouracil magnetic albu- min microspheres (5-Fu-MAMS) combined with external magnetic fields on human pancreatic carcinoma cells in vitro, and the mechanism. Methods The cultured PC-3 cells were divided into three groups: the group of 5-Fu-MAMS combined with external magnetic fields ; the group of 5-Fu-MAMS without magnetic fields, and 5-Fu group. The growth of PC-3 cells were observed under an inverted microscopy, and the inhibitory rate (IR) was assayed by methyl thiazol tetrazolium (MTT) assay. Results The inhibitory rate of 5-Fu-MAMS group and 5-Fu group was 57.7% and 59. 6% respectively (P 〉 0. 05). The inhibitory rate of 5-Fu-MAMS combined with external fields was significantly increased to 60% (P 〈 0. 05 ). Conclusion There were same pharmacological action between 5-Fu-MAMS and 5-Fu. The growth inhibiting action was significantly increased after 5-Fu-MAMS combined with external magnetic fields.
