摘要
目的研究化湿液对湿阻证模型大鼠小肠粘膜损伤修复的影响。方法将50只SD大鼠随机分为正常组、模型组,化湿液低、中、高剂量组,每组10只。除正常组外,其余4组用改进后的环境加疲劳法制造湿阻证模型,连续造模6d。造模成功后,正常组和模型组大鼠按2mL/100g的剂量灌胃给予生理盐水;化湿液低、中、高剂量组分别按含生药0.4g/100g、0.8g/100g、1.6g/100g的剂量灌胃给予化湿液,1次/d,连续8d后取各组大鼠小肠组织,光镜下观察小肠粘膜损伤情况;用免疫组化法检测小肠SS、VIP及Fas蛋白的表达。结果与正常组比较,模型组大鼠小肠粘膜糜烂,绒毛肿胀,上皮变性、坏死、脱落,小肠SS、VIP及Fas的阳性反应均较多,染色强度显著升高(P<0.05);与模型组比较,化湿液治疗后湿阻证大鼠小肠结构清晰,粘膜完整,小肠SS、VIP及Fas的阳性反应均减少,染色强度显著降低(P<0.05)。结论化湿液对湿阻证大鼠小肠粘膜损伤具有修复作用,其机制可能与抑制小肠SS、VIP及Fas的异常表达有关。
Objective To study on the effects of Huashi liquid on repairing intestinal mucosal injury of rats with dampness retention syndrome.Method 50 SD rats were randomly divided into normal group,model group,high,middle and low-ose of Huashi liquid groups,10 rats in each group.Dampness retention syndrome was induced in all rats,except in normal group,by modified environmental and fatigued method for 6 days.After the mode was formed,the normal and model groups were fed with saline(NS) in 2 ml/100 g dose,high,middle and low-dose of Huashi lquid groups were fed with Huashi lquid in different dose(0.4g/100g,.8g/100g,.6g/100g),respectively,one time per day,and last for 8 days.Then,impaired extent of small intestinal mucosa was observed under light microscope and the expression of Somatostatin(SS),Vasoactive Intestinal Peptide(VIP) and Fascin(Fat) in small intestine were detected by immunohistochemistry.Results In small intestinal tract of model group,mucosal erosion,villus swelling,epithelium necrosis and shedding,the expression of SS,VIP and Fas significantly increased(P0.05) compared with the normal group.In small intestinal tract of all the Huashi liquid,clear structure and intact mucosathe expression of SS,VIP and Fas significantly decreased(P0.05) compared with the model group.Conclusions Huashi liquid could repair small intestinal mucosa impairment of rats with dampness retention,the mechanism may be related to regulating the expression of SS,VIP and Fas in small intestine.
