摘要
目的:探讨甲状腺素对阿尔茨海默病患者淋巴细胞糖原合酶激酶-3(glycogen synthase kinase-3,GSK-3)的影响。方法:用^(32)P标记法检测正常老年人组,阿尔茨海默病(Alzheimer's disease,AD)组,甲状腺素治疗组淋巴细胞糖原合酶激酶-3的变化,并用放射免疫法检测这些组血清三碘甲状腺原氨酸(triiodothyronine,T_3)、T_4甲状腺素(tetraiodothyronine,T_4)、游离三碘甲状腺原氨酸(freetriiodothyronine,FT_3)、T_4游离甲状腺素(free thyroxine,FT_4)、促甲状腺激素(thyrotropin,TSH)的变化。结果:与对照组比较,AD组及甲状腺素组治疗前T_3及FT_3的含量存在明显降低(P<0.01,P<0.01)。TSH水平也明显降低(P<0.01,P<0.01),且伴有淋巴细胞GSK-3活性的明显降低(P<0.01,P<0.01)。甲状腺素治疗后,T_3、FT_3、TSH水平明显恢复(P<0.01),淋巴细胞GSK-3的活性明显降低(P<0.01),与对照组无明显差别(P>0.05)。结论:AD患者甲状腺功能降低,且伴有GSK-3活性升高,甲状腺素治疗后甲状腺功能恢复,且GSK-3活性降低,提示甲状腺素对AD治疗可能有价值。
Objective:our pruepose was to explore the effects of thyroid on glycogen synthase kinase-3activity in lymphocytes in alzheimer disease.methods:the activity of GSK-3 was measured ty 32^p liquild scintillography for incorporated radioactivity,and the levels of serum triiodothy romine(T3) ,teraiodothyronine(T4),free triiodotyronine(FT1),free thyroxine(FT4),and thyrotropin(TSH)were measured by radioimmunoassay in control group.Alzheimer disease group and thyroid treament group.Results:Compared with control group,the levels of T3、FT3and TSH were lower in AD group and in thyroid group unpretreatment(P〈0.01,P〈0.01),moreover,GSK-3 activity was also increased in these two groups(P〈0.01,P〈0.01),The higher levels of T3,FT3 and TSH were induced in thyroid treatment grop(P〈0.01),and the activity of GSK-3 activity in AD group,thyroid function was recoveried and low level of GSK-3 activity was reversed after thyroid treatment. Out results suggust that thyroid be valuble for AD treatment.
