apoB基因EcoRI、XbaI、MspI及apoAI基因-75bp、+83bp位点多态性与哈萨克族冠心病的相关性 被引量:3

DNA polymorphisms of the apolipoprotein B and AI gene in patients with coronary artery disease in Kazaks

摘要 目的:分析载脂蛋白B(apoB)基因EcoRI、XbaI、MspI位点和载脂蛋白AI(apoAI)基因-75bp、+83bp位点多态性与哈萨克族人冠心病的关系。方法:采用聚合酶链式反应-限制性片段长度多态性分析法检测185例哈萨克族冠心病患者和216例哈萨克族非冠心病对照者的这些位点多态性。结果:①TC、TG及apoAI/apoB在2组之间存在显著差异。②各个位点各基因型及等位基因频率在2组之间无显著差异。E++/X+-/Ms+-/M1+-/M2++(基因型联合10)频率在冠心病组和对照组的比值(12.9%∶6.5%)差异有统计学意义(P<0.05)。③冠心病组中基因型联合10的患者TC水平显著高于本组中其余基因型联合总体(基因型联合1~9)的平均水平,apoAI/apoB水平显著低于本组中基因型联合1~9的平均水平;对照组中基因型联合10的apoAI/apoB水平也显著低于本组中基因型联合1~9。④冠心病组基因型联合2的apoAI/apoB显著高于同组基因型联合10,对照组也是如此,而且对照组基因型联合2的LDL-C显著低于同组基因型联合10。⑤冠心病组基因型联合4的TC水平显著高于本组基因型联合2,其apoAI/apoB显著低于基因型联合2;对照组基因型联合4的apoAI/apoB显著低于同组基因型联合2及10,LDL-C显著高于基因型联合2。结论:基因型联合10与哈萨克族人冠心病相关。其中的机制可能与载脂蛋白基因变异引起apoAI/apoB的改变进而引起TC、TG等的变化有关。基因型联合4可能也是冠心病的易患因素,基因型联合2可能是冠心病的保护因素。 Objective:To explore the relationship between polymorphisms of EcoRI,XbaI and MspI of apolipoprotein B(apoB) gene and-75bp,+83bp of apolipoprotein AI(apoAI) gene and coronary heart disease(CHD) in Kazaks of Xinjiang,China.Method:These polymorphisms were analyzed by PCR-restriction fragment length polymorphism(PCR-PFLP).185 patients with CHD and 216 controls were involved.Result:①There were significant differences in TC、TG、apoAI/apoB and hypertension between the two groups.②Polymorphisms of the five sites were not related with CHD respectively.The polymorphism coalition frequency of E+-/X——/Ms++/M1+-/M2++(named Coalition 10) is significantly higher in CHD compared with control group(21.8% vs 10.4%,P0.05).③Coalition 10 in CHD is associated with increased TC and decreased apoAI/apoB compared with the other polymorphism coalitions as an unit(Coalition 1~9) in the same group significantly.Coalition 10 in control group is also associated with decreased apoAI/apoB compared with Coalition 1~9.④The level of apoAI/apoB of Coalition 2 in CHD group is significantly higher compared with it in Coalition 10 as it in control group.The level of LDL-C of Coalition 2 is significantly lower than it in Coalition 10 in control group.⑤TC of Coalition4 is significantly higher and apoAI/apoB of Coalition 4 is significantly lower than them in Coalition 2 in the CHD group.LDL-C of Coalition 4 is significantly higher than it in Coalition 2,and apoAI/apoB of Coalition 4 is significantly lower than it in Coalition 2 or 10 in the contnol group.⑥ApoAI/apoB is negatively related with TC and is positively related with HDL-C both in CHD and control groups.Conclusion:Coalition 10 of apoB and apoAI gene polymorphisms is correlated with CHD in Kazaks,and perhaps this phenomenon is begin with decreased apoAI/apoB.Coalition 4 may also be associated with CHD,and Coalition 2 may be a protective factor for CHD.
