RANK在大鼠股骨干骺端组织上的表达及其意义

摘要 目的检测大鼠股骨干骺端RANK基因的表达及其意义。方法取36只Wistar雌性大鼠,随机分为实验组(OVX)组和对照组(Sham-OVX),实验组无菌条件下腹侧入路行完整双侧卵巢摘除;对照组摘除与卵巢重量相同的卵巢周围脂肪组织各1块。术后均常规喂食3w。多聚甲醛主动脉灌注固定半小时后取出股骨干,多聚甲醛液固定24h,EDTA溶液中脱钙4~6w后,应用免疫组织化学染色的方法检测不同鼠龄的去势大鼠股骨干骺端中RANK的表达。结果在老年雌性大鼠的股骨干骺端皮质骨外膜中RANK的表达较12w鼠龄的股骨干骺端中RANK的表达减弱,骨内膜中RANK的表达在不同的鼠龄中变化不明显。在去势大鼠的小梁骨中RANK的表达较假手术组大鼠明显增强。结论雌激素缺乏可以增加大鼠股骨干骺端小梁骨中RANK的表达。 Objective To investigate the expression and significance of RANK in rat femoral metaphysis. Methods A total of 36 male Wistar rats were randomly divided into ovariectomized group (OVX) and sham-operated group (Sham-OVX). Rats in OVX group were performed bilateral ovariectomy under sterile condition. Fat tissue of the same weight of the ovary in adjacent region was cut from the rats in Sham-OVX group. The rats were fed routinely. After irrigation with paraformaldehyde for half an hour, the femoral shafts were collected. After 24h fixation with paraformaldehyde and 4-6w decalcification with EDTA, the expression of RANK in femoral metaphysis in rats of different age was examined using immunohistochemistry method. Results The RANK expression in periosteum of the femoral metaphysis in aged rat was less than that in 12w-old rats. The RANK expression in endosteum was constant in rats of different ages. The RANK expression in the trabecular bone increased more significantly in OVX rats than in Sham-OVX rats. Conclusion Estrogen deficiency results in an increase of RANK expression in the trabecular bone of femoral metaphysis in rats.
