摘要
目的探讨模拟失重对大鼠心肌细胞系-H9c2细胞的细胞形态、细胞骨架和增殖能力的影响。方法采用双向多样本回转器(2D-RWVs)模拟失重,在30 r/m in转速下分别培养2,4,6和8 d,用图像处理分析软件(Image J)测量分析模拟失重后细胞面积的改变,激光共聚焦荧光显微镜观察细胞骨架的变化,采用细胞计数法测量细胞生长曲线。结果回转组H9c2细胞在回转培养至8 d比对照组细胞面积显著缩小(P<0.001),细胞微丝和微管结构发生改变,改变率为48.48%,而回转培养2,4和6 d与对照组细胞面积、微丝和微管结构无明显差异。细胞生长曲线显示模拟失重8 d时间段,回转组H9c2细胞生长曲线与对照组一致。结论回转器模拟失重8 d导致H9c2细胞面积缩小、细胞微丝解聚以及微管结构紊乱,但其增殖能力未受到影响。
Objective To investigate the effects of simulated weightlessness on shape,cytoskeleton and proliferation of H9c2 cells.Methods H9c2 cells were cultured under simulated weightlessness with two-dimensional rotating wall vessel(2D-RWV),and under earth gravity as control for 2,4,6 and 8 d respectively.The changes of cell area were measured with image processing and analysis soft(image J).The cytoskeleton alteration was detected with laser confocal fluorescence microscopy.Cell growth curve was examined with cytometer.Results After 8 d simulated weightlessness,the area of H9c2 cells decreased significantly(P0.001) as compared with control;microfilament and microtubule tended to disassemble and showed derangement;cell growth curve revealed that cell proliferation was not inhibited.Conclusion Simulated weightlessness may lead to decrease in cell area and damage to cytoskeleton on day 8 when cultured in 2D-RWV,while no influence on the H9c2 cell proliferation.
