摘要
目的:探讨瘦素对脂肪细胞SOCS-3表达的影响及机制。方法:分离并培养大鼠成熟脂肪细胞,采用50 nmol.L-1重组瘦素对脂肪细胞分别处理0.5、2、6 h,免疫共沉淀结合Western blotting法检测SOCS-3、p-STAT1、p-STAT3蛋白表达。结果:瘦素未处理的脂肪细胞中有少量SOCS-3表达,在瘦素处理达6 h后,SOCS-3蛋白的表达量显著增加(P<0.01)。瘦素处理2 h和6 h后,p-STAT1蛋白表达相比于未处理和处理0.5 h的显著增加(P<0.01);p-STAT3蛋白表达在瘦素处理6 h后显著增加(P<0.01)。结论:瘦素可以通过激活Jak-STAT信号通路,诱导脂肪细胞SOCS-3的表达。
Objective: To study the effect and mechanism of leptin on suppressor of cytokine signaling 3(SOCS-3) expression in adipocytes.Methods: Mature adipocytes of rats were separated and cultured,and then they were incubated with 50 nmol·L-1 rat recombinant leptin for 0.5 h,2 h and 6 h,respectively.The expression of SOCS-3,p-STAT1 and p-STAT3 protein was detected by immunoprecipitation and Western blotting.Results: Low expression of SOCS-3 was detected in adipocytes without leptin treatment,and increased significantly after 6 h leptin treatment(P0.01).Compared to adipocytes without leptin treatment and with 0.5 h treatment,the expression of p-STAT1 increased markedly in adipocytes with 2 h and 6 h leptin treatment(P0.01),while the expression of p-STAT3 increased significantly only in 6 h leptin treatment(P0.01).Conclusion: Leptin can induce SOCS-3 expression through activating Jak-STAT signal pathway.
