摘要
目的研究曲格列酮对人胃癌细胞株BGC-823增殖和凋亡的影响。方法将不同浓度(0.5、1.0、5.0、15.0、25.0μmol/L)的曲格列酮分别作用于人胃癌细胞株BGC-823 48h后,通过免疫细胞化学测定胃癌细胞中增殖细胞核抗原(PCNA)表达情况,并通过原位末端标记(TUNEL)法测定其凋亡率。结果人胃癌细胞株BGC-823表达PCNA强阳性,经曲格列酮作用48h后,表达水平减弱。随曲格列酮浓度的递增,PCNA表达逐步递减。同时BGC-823凋亡率随药物浓度增加而明显递增。结论曲格列酮能够抑制人胃癌细胞株BGC-823增殖和诱导其凋亡,其效应具有浓度依赖性。
Objective To investigate the effects of troglitazone on proliferation and apoptosis in gastric cancer cell line BGC-823.Methods BGC-823 cells were treated with different concentrations(0.5,1.0,5.0,15.0 and 25.0 μmol/L)of troglitazone for 48 h,then the expression of proliferating cell nuclear antigen(PCNA) on the cells were detected by immunocytochemistry and the induction of apoptosis were investigated by TUNEL assay.Results The expression of PCNA could be observed in BGC-823 and down-regulated by troglitazone at the end of 48 h.The level of PCNA was gradually become weakened on the concentration range from 0.5 μmol/L to 25.0 μmol/L.At the same time,the apoptosis indexes of BGC-823 were significantly increased.Conclusion Troglitazone can inhibit the proliferation and induce apoptosis of the gastric cancer cell line BGC-823 in a dose dependent manner.
