广西古蓬种源马尾松遗传多样性的ISSR分析

Genetic diversity of Pinnus massoniana populations in Gupeng,Guangxi

利用ISSR分子标记技术对广西古蓬种源区3个不同地点的马尾松天然林中47个样品进行遗传多样性分析,从100个ISSR引物中共筛选出10个多态性引物,对这47个马尾松个体进行PCR-ISSR扩增,共检测到192个位点,其中多态位点数为177个。分析表明:在这3个马尾松天然林群体内,总的多态位点百分率P为94.35%,总的Shannon信息指数I为0.427 8,Nei’s基因多样度指数h为0.276 5;每个群体遗传多样度水平稍有不同,P平均值为86.63%,I平均为0.356 8,h平均为0.311 4,表明种水平的遗传多样性较高。马尾松组间的基因分化系数Gst为0.048 1,表明马尾松的主要变异存在于组间,3个组的基因流为9.887 7,表明基因流不是引起马尾松组间分化的主要因素。

Genetic diversity of Pinus massoniana Lamb.（mass pine） populations were evaluated using inter simple sequence repeat（ISSR） molecular technique.Amplification was conducted with 10 primers for 47 individuals selected from 3 populations.A total of 177 polymorphic and 15 shared markers were detected.The percentage of polymorphic marker P was 94.359%.The Shannon＇s information index I and Nei＇s gene diversity h were 0.472 8 and 0.276 5,respectively,which indicated a high genetic diversity at species level.The mean values of P,I and h were 86.63%,0.356 8 and 0.311 4.Gene differentiation coefficient among groups was 0.048 1 and gene flow of three groups was 9.887 7,which concluded that gene flow was not the factor for genetic differentiation among groups.
