摘要
黑柄炭角菌发酵菌丝经热水浸提、Sevag法脱蛋白、透析、乙醇沉淀、DEAE-52纤维素层析和SephadexG-100凝胶层析等分离纯化,得到黑柄炭角菌多糖组分XNW-1,紫外扫描检测和Sephadex G-100分析表明XNW-1为均一组分,相对分子质量9.0×105.单糖组成分析表明XNW-1只含葡萄糖.XNW-1的糖含量为96%.红外光谱、GC-MS、1H和13C核磁共振结构分析表明:XNW-1由α型糖苷键构成,(1→4)葡萄糖(82%)的连接方式,构成主链的核心;其他连接方式包括(1→4,6)葡萄糖(12%),(1→6)葡萄糖(1%),末端(1→)连接葡萄糖(5%);末端糖基所占比重不大,说明XNW-1为低分支结构.
The polysaccharide XNW-1 was isolated from the fermented mycelium of Xylaria nigripes(Klotzsch) Sacc.through hot-water extraction and subjected to Sevag deprotein,dialysis,ethanol precipitation,DEAE-52 cellulose ion-exchange column chromatography and Sephadex G-100 column chromatography.This polysaccharide was found to be highly homogeneous with a relative molecular mass of 9.0×105.Analysis of monosaccharide composition revealed XNW-1 were only made up of glucose.The total sugar content of XNW-1were 96%.The chemical structure of XNW-1 was analyzed by IR,GC-MS,1H-NMR,and 13C-NMR.These results indicated that XNW-1 has the following structure: XNW-1 is mainly made up of α configuration glycosidic,its basic structure was composed of major(1→4)linked glucose(82%),other linkages including(1→4,6)glucose(12%),(1→6)glucose(1%) and terminal(1→)glucose(5%).XNW-1 was a kind of lowly branched structure.
