摘要
建立了快速、稳定的家兔血浆中地克珠利提取方法,为进一步研究家兔组织中地克珠利的残留量提供基础。采用HPLC法以妥曲珠利为内标,N,N-2甲基甲酰胺及乙腈联合沉淀法提取家兔血浆中地克珠利,色谱柱为Hypersil ODS(2.5μm,250 mm×4.6 mm),流动相为乙腈∶水∶三乙胺∶冰乙酸=55∶45∶0.2∶0.12(V/V),流速为1.0 mL/min,紫外检测波长为240 nm。在0.156~20.0μg/mL浓度范围内,地克珠利的峰面积与其浓度呈现良好的线性关系(r2=0.999 2);回收率为98.9%~102.4%;方法精密度RSD最大值为6.8%;最低检测限为0.078μg/mL。DMF及乙腈联合沉淀法适用于家兔血浆中的地克珠利的提取。
To establish a rapid and stable tissue extraction method for the determination of diclazuril residues in rabbit plasma,diclazuril and toltrazuril(internal standard)were added in blank rabbit plasma,extracted by N,N-dimethylformamide(DMF)/acetonitrile precipitation method and determined by HPLC(Hypersil ODS-C18).The mobile phase consisted of a mixture of tirethylamine/water/triethylamine/aceticacid(55∶ 45∶ 0.2∶ 0.12) at the flow rate was 1.0 mL/min.The ultraviolet detection wavelength was set at 240 nm.The method was proven to be linear over a plasma diclazuril concentration range of 0.156 to 20.0 μg/mL with a mean correlation coefficient of 0.999 2.The recovery of diclazuril from rabbit plasma was found to be 98.9%~102.4%.The max RSD of method precision was found to be 6.8%.The detection limit was 0.078 μg/mL.DMF/acetonitrile precipitation method is suitable for the extraction of diclazuril in rabbit plasma.
