摘要
用液体培养的方法,研究培养基、pH值、温度以及氧气等因素对北美大豆猝死综合症病菌(Fusarium virgu-forme sp.nov.)菌株F-171厚垣孢子产生的影响。结果表明:菌株F-171产生厚垣孢子的最适液体培养条件为7 500 r.min-1、2 min匀浆断裂菌丝,在250 mL三角瓶装入80 mL大豆茎秆或豆芽汁培养液,在pH 6,25~30℃下,以120 r.min-1振荡培养。最佳液体培养条件下的产孢曲线表明,随培养时间的延长,厚垣孢子产量逐渐增加,10 d后产量最高,达到3.15×105个.mL-1。
The effects of culture medium,pH,temperature and oxygen on the chlamydospore production of Fusarium virguliforme sp.nov.strain F-171 were studied by liquid culture method.The results showed that,the best liquid culture conditions for the chlamydospore production of the strain F-171 were,homogenized hyphae at 7500 r·min-1 for 2 min,filled 80 mL liquid culture of soybean stem or sprout juice in the 250 mL flask,and incubated at pH 6,25-30℃ and 120 r·min-1 shaking culitivation environment.Chlamydospore production curve in the best liquid culture conditions showed that the number of chlamydospore increased gradually with prolonging of the incubation time,reached the peak(3.15×105spores·mL-1)10 days after incubation.
