摘要
建立了快速测定大麦籽粒中13种酚酸类化合物(没食子酸、原儿茶酸、绿原酸、对羟基苯甲酸、咖啡酸、香草酸、丁香酸、间羟基苯甲酸、对香豆酸、阿魏酸、藜芦酸、邻香豆酸和水杨酸)的反相高效液相色谱方法。采用SUPELCO AscentisC18色谱柱(150 mm×4.6 mm,5μm)分离,流动相为甲醇-0.1%甲酸水溶液,梯度洗脱,流速为0.7 mL/min,柱温为40℃;检测波长为275nm,进样量为10μL,20 min内13种组分得到了较好的分离。各组分的质量浓度与其峰面积有良好的线性关系,相关系数均>0.999。13种组分的检出限为0.30~5.0mg/L,平均回收率为87.0%~105.5%,相对标准偏差为0.04%~3.1%。方法可用于大麦籽粒中酚酸类化合物的快速测定和评价。
A reversed phase high performance liquid chromatographic method for the rapid determination of 13 phenolic acids (including gallic acid, protocatechuic acid, chlorogenic acid, p-hydroxybenzoic acid, caffeic acid, vanillic acid, syringic acid, m-hydroxybenzoic coumaric acid, and salicylic acid) in barley grain was Ascentis C18 column (150 mm ×4. 6 mm,5 μm) at acid, p-coumaric acid, ferulic acid, veratric acid, odeveloped. The separation was performed on a SUPELCO 40℃ by gradient elution. The mobile phase consisted of methanol and 0. 1% formic acid solution at a flow rate of 0. 7 mL/min. The injection volume was 10 μL, and the detection wavelength was 275nm. All of the phenolic acids were weU separated within 20 rain. There were good linear correlations between the concentrations and peak areas of the 13 components with the correlation coefficients all above 0. 999. The detection limits of the components were 0. 30 - 4.99 rag/L, and the average recoveries were 87.09% - 105.5% with relative standard deviations of 0. 04% - 3.1%. Good repetition and precision of the method were demonstrated through the results, and it can be applied to the rapid determination and evaluation of phenolic acids in barley grain.
