犬口服盐酸特比萘芬片的生物利用度研究 被引量:4

摘要 将12只健康犬随机分为2组,按照随机2×2交叉法设计,分别按单剂量(30 mg/kg)静注盐酸特比萘芬溶液、口服盐酸特比萘芬片,用反相高效液相色谱法测定犬血浆中盐酸特比萘芬的浓度,3P97药动学计算软件处理血浆药物浓度-时间数据。结果显示,静脉注射盐酸特比萘芬在犬体内药动学过程符合无吸收开放二室模型,主要药物代谢动力学参数为:T1/2α(0.37±0.07)h,T1/2β(5.57±5.15)h,V(1.27±0.19)L/kg,CL(1.38±0.34)L/h,AUC(21.85±6.63)μg/(h.mL);口服盐酸特比萘芬符合一级吸收一室模型,主要药动学参数为:T1/2Ka(2.55±0.51)h,T1/2Ke(4.84±0.92)h,V/F(17.23±2.68)L/kg,Tm ax(4.95±0.82)h,Cm ax(0.86±0.08)μg/mL,AUC(13.66±2.77)μg/(h.mL)。口服盐酸特比萘芬片剂的生物利用度为55.53%。表明盐酸特比萘芬片剂在健康犬体内吸收较快,有效血药浓度持续时间长,生物利用度较高。
