摘要
目的:测定七叶莲提取物中总黄酮及槲皮素的含量。方法:总黄酮含量测定,以芦丁为对照品,采用分光光度法,于509nm处测定;槲皮素含量测定,采用高效液相色谱法,色谱柱为Agilent ZORBAXSB-C18(4.6 mm×250 mm,5!m),流动相为乙腈-0.4%磷酸溶液(36∶64),流速1.0 mL.min-1,检测波长为254 nm,柱温25℃。结果:总黄酮线性范围为0.0~40.0μg.mL-1(r=0.999 9),槲皮素线性范围为0.06~2.40!g(r=0.999 2);总黄酮和槲皮素的平均回收率分别为100.6%和98.7%。结论:建立的方法具有良好的精密度和耐用性,结果准确可靠,可作为本产品的质量控制方法。
Objective: To determine the contents of total flavonoids and quercetin in Schefflera venulosa extracts. Methods: The content of total flavonoids was determined with Vis - UV spectrophotometry;The wavelengh was set at 509 nm;Rutin was used as the control. The separation and quantitation of quercetin was performed on a Agilent ZORBAXSB - C18(4. 6 mm × 250 mm,5 μm) column with the mobile phase consisting of acetonitrile - 0. 4 % phosphoric acid(36: 64) at the flow rate of 1, 0 mL · min^-1 ,the detective wavelength wss 254 nm ,and the colunm temperature was 25 ℃. Results: The standard curve was linear in the range of 0. 0 - 40. 0 μg · mL^-1 for total flavonoids ( r = 0. 9999) and 0.06 - 2.40μg for qnercetin (r = 0. 9992), The average recoveries of total flavonoids and quercetin were 100. 6 % and 98.7 % ,respectively. Conclusions: The methods are simple,accurate and with a good reproducibility and can be used to control the quality of Schefflera venulosa extracts.
