Determination of rifampicin by flow-injection chemiluminescence

摘要 目的:建立一种流动注射-化学发光测定利福平的快速、灵敏新方法。方法:碱性条件下,利福平对luminol-KIO4化学发光体系有抑制作用,结合流动注射技术,建立了流动注射-化学发光检测利福平的新方法。结果:在优化条件下,利福平的线性范围是7.0×10-9~6.0×10-7 mol.L-1,检出限达3.2×10-9 mol.L-1。对3.0×10-8 mol.L-1和2.0×10-7mol.L-1利福平分别进行11次测定,相对标准偏差分别为0.8%和1.6%。分析速度为128个样品/时。将提出的新方法成功应用于利福平滴眼液和利福平胶囊中利福平含量的测定。结论:本方法操作简单快速,适合于利福平的分析。 Objective : To establish a novel, rapid and sensitive method for the determination of rifampicin (RFP) using flow - injection (FI) coupled with chemiluminescence (CL) detection. Methods : RFP was found to be inhibit the CL signal of the luminol - KIO4 CL system in alkaline medium. Therefore, a new FI - CL method was developed for the determination of RFP. Results: Under the optimum conditions, the net CL intensity was linear with the RFP concentration in the range of 7. 0×10^-9 ~ 6.0 ×10^-7 mol · L^-1 with the detection limit of 3.2 ×10^-9 mol · L^-1. The relative standard deviations (RSDs)were found to be 0. 8% and 1.6% for 11 repeated determinations of 3.0 ×10^-8 mol · L^-1 and 2. 0 ×10^-7 mol · L^-1 RFP,respectively. The analyzing rate is about 128 samples/h. The proposed method has been successfully applied for the determination of RFP in RFP for eye use and RFP capsules. Conclusions:The method is convenient,rapid, accurate and may be considered for the analysis of RFP.
