孕酮对大鼠大脑皮质神经元氨基酸受体亚单位GBR1和NR2B表达的影响

Effects of progesterone on amino acids receptor subunit GBR1 and NR2B expression in primary cultured rat cerebral cortical neurons in vitro

摘要 目的:研究孕酮(PROG)对大鼠大脑皮质神经元氨基酸受体亚单位GBR1和NR2B表达的影响。方法:不同剂量(1,10,100μmol.L-1)的孕酮处理原代培养的大鼠大脑皮质神经元,采用免疫细胞化学法测定神经元氨基酸受体亚单位GBR1和NR2B表达的变化。结果:与对照组相比,1,10,100μmol.L-1 PROG使GBR1表达分别增加45.9%、172.0%、211.6%(P<0.01),使NR2B表达分别增加42.1%、51.4%、88.4%(P<0.05或P<0.01)。结论:孕酮可增强氨基酸受体亚单位GBR1和NR2B的表达水平。 OBJECTIVE To investigate the effect of progesterone(PROG) on the levels of GBR1 and NR2B expression in primary cultured rat cerebral cortical neurons. METHODS Primary cultured rat cerebral cortical neurons were treated with different concentrations PROG (1, 10, 100 μmol. L-1 ) and the expression of amino acids receptor subunit GBR1 and NR2B were detected by immunocytochemistry. RESULTS Compared with control group, the average optical density of GBRI increased by 45.9%, 172. 0%,211.6% after incubating with PROG,PROG-M,PROG-H (P〈0. 01), the average optical density of NR2B increased by 42. 1%,51.4%,88. 4% after PROG-L,PROG-M,PROG-H treatment(P〈0. 05 or P〈0. 01 )as well. CONCLUSION PROG can increase the expression of neuron receptor subunit GBR1 and NR2B.
