摘要
目的探讨丁香胃灵合剂(CWLM)对慢性萎缩性胃炎(CAG)大鼠生长抑素(SS)、Bcl-2、Bax的影响及机制。方法将60只健康Wistar大鼠随机分为5组,即正常组,CAG组,CWLM低、中、高剂量组,每组12只。采用水杨酸钠联合氨水建立大鼠CAG模型,造模2月后开始给予药物干预,CWLM按16,8,4 g/(kg.d)的剂量给予大鼠连续灌胃50 d,末次给药1 h后,采用ELISA法检测血清SS的含量,免疫组化法检测胃黏膜中Bcl-2和Bax蛋白的表达变化。结果与正常组比较,CAG组血清SS含量和胃黏膜Bcl-2蛋白阳性表达率显著升高,Bax蛋白阳性表达率显著降低(P<0.01);与CAG组比较,CWLM高、中剂量组血清SS含量显著降低,Bax蛋白阳性表达率显著升高,CWLM高剂量组Bcl-2蛋白阳性表达率显著降低(P<0.01),其中高剂量组作用最为显著(P<0.01或P<0.05)。结论 CWLM对CAG大鼠模型有较好的治疗作用,而减少血清SS含量、下调胃黏膜Bcl-2蛋白表达或上调Bax蛋白表达可能是其重要的作用机制之一。
Objective To explore the effects and mechanism of Clove Weiling Mixture(CWLM) on somatostatin(SS),Bcl-2 and Bax in rats with chronic atrophic gastritis(CAG).Methods 60 Wistar rats were randomly divided into normal group,CAG group,CWLM low-dose group,CWLM middle-dose group and CWLM high-dose group,each group had 12 rats.CAG model was established in rats by sodium salicylate and ammonia water.Drug intervention began after being made model 2 months.The rats were treated with 16,8,4 g/(kg·d) dose of CWLM.After 50 days,all rats were anesthetized to measure the content of SS in blood serum by ELISA method,and the expression of Bcl-2 and Bax in gastric mucosa by immunohistochemistry method.Results Compared with normal group,the content of SS in blood serum and the positive expression rate of Bcl-2 in gastric mucosa in CAG model were significantly increased,but the positive expression rate of Bax was significantly decreased(P〈0.01).Compared with CAG group,the content of SS of CWLM middle-dose group and CWLM high-dose group were significantly decreased and the positive expression rate of Bax was significantly increased,the positive expression rate of Bcl-2 of CWLM high-dose group was significantly decreased(P〈0.01),and the effects of CWLM high-dose group were most significant(P〈0.01 or P〈0.05).Conclusion CWLM has better curative effects on CAG rat model,and decreasing the content of SS in blood serum and the expression of Bcl-2 in gastric mucosa,increasing the expression of Bax may be one of mechanisms of its action.
