紫杉醇、硒酸酯多糖序贯给药对乳腺癌MCF-7细胞抑制作用的研究 被引量:1

摘要 目的:研究硒酸酯多糖与紫杉醇单独及序贯给药对体外培养乳腺癌MCF-7细胞的生长抑制作用及对细胞周期的影响。方法:采用MTT法及SRB法检测药物对乳腺癌MCF-7细胞体外杀伤作用,应用金氏公式判断联合用药效果,流式细胞仪检测细胞周期及凋亡率。结果:MTT及SRB法结果显示,药物对MCF-7细胞的抑制作用呈剂量依赖性,两者联合作用效果优于单独作用组,30mg/L的硒酸酯多糖与0.5~2nmol/L紫杉醇联合应用表现为协同作用,与4nmol/L紫杉醇联合应用表现为相加作用。流式细胞术结果显示,紫杉醇作用于G1期,同时添加组及序贯给药组既可发生S期和G2/M期周期阻滞,又可诱导凋亡,但凋亡率低于Taxol单独组。结论:紫杉醇与硒酸酯多糖对乳腺癌MCF-7细胞有协同抑制增殖和诱导凋亡的作用。 Objective:To investigate the inhibitory effect of paclitaxel(taxol) combined with k-selenocarrageenan(KSC) on growth of breast cancer MCF-7 cell. Methods:The inhibitory effects of the drugs on the growth of breast cancer cells were determined by MTT assay and Sulforhodamine B(SRB).The Jin's formula was used to analyze the effect of combined drug therapy.Cell cycle distribution and apoptosis rate were assessed by flow cytometry(FCM). Results:taxol and KSC have obvious growth inhibiting effect on MCF-7 cells in a dose-dependent manner.They showed the effect of the joint application was much stronger than respective application.Co-administration of 30mg/L KSC and 0.5~2 nmol/L taxol had synergistic inhibitory effects on the proliferation.Co-administration of 30mg/L KSC and 4 nmol/L taxol had additive inhibitory effect.FCM results demonstrated that taxol made MCF-7 cells G1 phase arrest.G2/M and S phase block and apoptotic peak were appeared in the combination groups,but the apoptotic rates were lower than that in the Taxol alone. Conclusion:taxol and KSC can inhibit the proliferation of MCF-7 cells and induces apoptosis.
