多聚乙酰亚胺介导CNP基因对兔髂动脉球囊成形术后内膜增生的影响

摘要 目的观察局部转染C型利钠肽基因(CNP)对血管内膜增生的影响,探讨CNP在防治球囊成形术后再狭窄中的作用.方法构建兔髂动脉粥样硬化模型,将多聚乙酰亚胺-pcDNA3.1/hCNP包被球囊转染髂动脉,分别用RT-PCR、免疫组化、Western blot等方法检测pcDNA3.1/hCNP在动脉壁中的表达,弹力纤维染色法检测球囊成形术后内膜增生等指标.结果 pcDNA3.1/hCNP在兔髂动脉中能高效表达,转染组血管内膜增生程度明显降低.结论 CNP基因转染能明显抑制血管内膜增生,在血管再狭窄防治中可能具有应用价值. Objective To discuss the potential role of C-type natriuretic peptide(CNP)in vascular restenosis prevention after aortic sac plasty by observating the influence of CNP on the intima hyperplasia.Methods Reconstructed plasmid pcDNA3.1/hCNP was transfected with polyethylenimine(PEI)by catheter method in iliac artery of rabbit model of atherosclerosis.The expression of pcDNA3.1/hCNP was confirmed with RT-PCR,immunohistochemistry and Western blot.The hyperplasia of intima was detected with elastic fibers staining.Result It was confirmed that the reconstructed pcDNA3.1(+)/hCNP was expressed effectively in vivo with markedly reduced hyperplasia of intima.Conclusion CNP gene transfection can inhibit the hyperplasia of intima and has potential application value in prevention of vascular restenosis.
