摘要
目的:探讨氧化苦参碱对银屑病模型雌激素周期小鼠阴道上皮细胞增殖和凋亡的影响。方法:采用注射雌激素刺激小鼠阴道上皮组织增生的方法复制银屑病小鼠动物模型,同时给予不同剂量的氧化苦参碱干预,实验10 d后,应用免疫组化法和末端转移酶介导的缺口末端标记(TUNEL)技术分别检测各组小鼠阴道上皮细胞细胞增殖性核抗原(PCNA)表达水平和凋亡情况。结果:与正常组相比,模型组PCNA表达显著增加(P<0.01),细胞凋亡指数明显增加(P<0.01),氧化苦参碱80、40和20mg/kg组PCNA表达显著减少(P<0.01),凋亡指数显著增加(P<0.01)。结论:氧化苦参碱对实验性银屑病具有保护作用,该作用可能与其降低表皮细胞PCNA表达水平,促进表皮细胞凋亡有关。
Objective: To investigate the effect of oxymatrine on the expression levels of PCNA and apoptosis in psoriasislike mice model. Methods: The mouse vaginal epithelia in estrus cycle were used to establish psoriasis-like model by injecting estrogen, and then the mouse were treated with three doses of oxymatrine. The action of oxymatrine in regulating the expression of PCNA and apoptosis index was observed. Results: Compared with the normal group, the expression of PCNA and the apoptosis index of vaginal epithelial cells increased markedly in model group (P 〈 0.01). Oxymatrine could significantly reduce the expression of PCNA (P〈 0.01) and increase the apoptosis index significantly (P 〈 0.01). Conclusion: Oxymatrinc has good efficacy in treating animal psoriasis model by reducing the expression of PCNA and increasing the apoptosis index.
