胰腺癌EGFR和HER3的蛋白表达与基因状态分析

摘要 目的探索胰腺癌EGFR和HER3的基因状态、蛋白表达与胰腺癌临床病理因素之间的关系,分析它们之间的相关性,探讨EGFR和HER3在胰腺癌中的作用。方法分别采用荧光原位杂交(FISH)和免疫组织化学(IHC)方法,检测55例中国人胰腺癌及癌旁组织EGFR和HER3基因状态和蛋白表达,结合临床病理资料进行统计学分析。结果 55例胰腺癌中,EGFR(FISH)阳性33例(60.0%)。EGFR(IHC)阳性34例(61.8%)。HER3(FISH)阳性26例(47.3%)。HER3(IHC)阳性4例(7.3%)。统计结果显示,EGFR、HER3蛋白表达与基因扩增间无明显相关性(P>0.05)。EGFR与HER3之间在蛋白的表达以及基因状态上也无明显相关性(P>0.05)。EGFR、HER3蛋白表达及基因扩增与患者的性别、年龄、肿瘤大小、肿瘤分化程度、胰周浸润,肝转移均无关(P>0.05)。结论在胰腺癌中,EGFR的蛋白表达显著高于癌旁组织;HER3蛋白表达与癌旁组织无显著差异。胰腺癌中存在较高比例的EGFR、HER3基因扩增。EGFR、HER3蛋白表达与基因拷贝数增加无关。EGFR与HER3相互之间在蛋白表达及基因状态上也不相关。EGFR与HER3在胰腺癌中的作用尚不明确。 Objective To investigate the gene copy number and protein expression of EGFR and HER3 in pancreatic cancer and to determine the relationship between these markers and the pathological features of pancreatic cancer.Methods 55 cases of paraffin-embedded pancreatic cancer tissues and corresponding tumor adjacent pancreatic tissues from Chinese patients were analyzed by fiuorescence in situ hybridization(FISH) and immunohistochemistry(IHC) for gene copy number and protein expression,respectively.Results Among all 55 samples,pancreatic cancer EGFR FISH positive,in the form of high polysomy and gene amplification,were present in 33/55(60%) of patients,whereas EGFR protein were over expressed in 34/55(61.8%) detected by IHC.HER3 increased copy number were present in 26/55(47.3%),and the elevated protein level were diplayed in 4/55(7.3%).There was no correlation between gene copy or protein expression of EGFR and HER3(P0.05).No significant association between EGFR copy number and its corresponding protein level was found(P0.05),nor did HER3.The relationship between EGFR and HER3 with clinical pathological features of pancreatic cancer were not statistically significant(P0.05).Conclusion Increased EGFR,HER3 gene copy number and EGFR overexpression are present in a significant proportion of pancreatic cancer patients,but the expression of HER3 is not different from tumor adjacent pancreatic tissue.Neither EGFR nor HER3 is related to clinical pathological features of pancreatic cancer.There is no relationship between EGFR and HER3.There is no significant correlation between EGFR and HER3 expression and their corresponding gene copy number.The function of EGFR and HER3 remains unclear in pancreatic cancer and it needs more studies.
