摘要
目的:探讨Rho激酶抑制剂对体外培养的膀胱癌细胞侵袭能力的影响。方法:培养膀胱癌细胞株T24,采用CCK-8实验、体外细胞侵袭实验、细胞运动实验测Rho激酶抑制剂对细胞生长和侵袭运动能力的影响。结果:经Rho激酶抑制剂处理后的膀胱癌细胞株T24的侵袭和运动能力明显下降,呈浓度依赖性,且当药物浓度>75μmol/L后,细胞的生长受到明显的抑制。结论:Rho激酶信号转导系统对膀胱癌细胞的侵袭和生长有重要的影响。
Objective: To probe the effect of Rho kinase inhibitor on the invasive ability of bladder cancer.Methods: bladder cancer cell line T24 was cultured,CCK-8 experiments,in vitro invasion assay and cell movement experiment were used to measure the changes of the growth,movement and the invasion ability of bladder cancer when Rho kinase was inhibited before and after.Results: After treatment with Rho kinase inhibitor,the growth,invasion and movement of bladder cancer cells line T24 were inhibited obviously,and these were decreasing along with the increasing inhibited degree of the Rho kinase.And when the drug concentration 75 umol / L,the cell growth was inhibited obviously.Conclusion: Rho kinase signaling pathway importantly affects the cell invasion and growth of bladder cancer.
