摘要 采用盐析法对小刺猴头菌寡糖进行纯化,在pH值、硫酸铵饱和度和溶液浓度3个单因素试验的基础上,根据中心组合(Box-Benhnken)试验设计原理,以粗多糖中的蛋白去除率为指标响应值作响应面和等高线,分析各个因素的显著性和交互作用,得出利用硫酸铵盐析法去除寡糖中杂蛋白的工艺条件为:pH值为5.91,硫酸铵饱和度为57%,溶液浓度为4%。 The experiment was conducted using salting - out method to purify the oligosaccharide of H. caput - medusae ( Bull. Fr. ) Pers.. The pH, the saturation of ammonium sulfate and the solution concentration were investigated using single - factor experiment. Then the purification conditions of oligosaccharide of H. caput - medusae ( Bull. Fr. ) Pers. on the rationale of BoxBenhnken design of Response Surface Methodology experiment were optimized, and the response surface and contour were drawn out with the removal rate of protein in crude oligosaceharide content as criteria. Through the analysis of the distinctiveness and interactions between various factors, we can conclude that the conditions for eliminating proteins from oligosaccaride by saltingout method are that pH value is 5.91, the saturation of ammonium sulfate is 57%, and the solution concentration is 4 %.
