摘要
目的:观察鼠白介素28重组腺病毒(r-Ad-mIFN-λ2)感染小鼠后的表达。方法:将已成功构建的重组腺病毒r-Ad-mIFN-λ2感染小鼠,采用实时PCR、蛋白质印迹方法,检测小鼠骨骼肌mIFN-λ2基因和蛋白的表达,并监测小鼠的生长情况。结果:重组腺病毒Ad-mIFN-λ2在小鼠体内能够稳定表达,且不影响其正常生长。实时PCR结果显示Ad-mIFN-λ2组持续表达mIFN-λ2 mRNA,表达量高于空白组和空载体LacZ组;蛋白质印迹结果表明Ad-mIFN-λ2组出现明显mIFN-λ2蛋白的条带,空白组及LacZ组条带较淡;监测小鼠体质量表明感染r-Ad-mIFN-λ2的小鼠能够正常生长,与空白组和LacZ组之间差异无统计学意义。结论:构建的重组腺病毒Ad-mIFN-λ2在小鼠体内感染后能够稳定表达,且不影响其正常生长,为进一步研究mIFN-λ2在动物体内的生物学活性奠定了一定的基础。
Objective: To study the mil-28 expression in the rat transfected with the interleukin 28 recombinant adenovirus vector.Methods: Transfect the recombinant mil-28 adenovirus vector into the rat,detect the mil-28 gene expression with real-time PCR and Western blot.Results: The interleukin 28 recombinant adenovirus vector in the rat can express stably,and at the same time the rat can grow normally.In the muscle,the Ad-mIFN-lambda 2 group express the mIFN-lambda 2 gene at a higher level,while the blank group and the LacZ group at a lower level by the real-time PCR;Detected with the Western blot,Ad-mIFN-lambda 2 group appeared obviously about mIFN-lambda 2 protein strip,while the blank group and the LacZ group with lighter bands.Conclusion: The stable expression of the mIFN-lambda 2 gene in the rat was detected after transfected with the interleukin 28 recombinant adenovirus vector,and the rat can grow normally,which give a guidance to the vivo biological activity analysis of the mIFN-lambda 2 in the future.
