摘要
目的:揭示瘦素是否调控培养的肝星状细胞中固醇调节元件结合蛋白-1c(SREBP-1c)的表达。方法:用瘦素孵育培养的肝星状细胞,分别通过Western Blot和real-time PCR检测SREBP-1c的蛋白及其mRNA水平,并通过双荧光素酶报告基因检测SREBP-1c启动子活性。结果:瘦素明显抑制SREBP-1c的蛋白表达,mRNA的转录,瘦素还能抑制SREBP-1c基因的启动子水平。结论:揭示了瘦素在肝纤维化发生中具有独特促进作用。
Objective: The aim of this study is to examine whether leptin regulates SREBP-1c gene expression in HSCs in vitro.Methods: Cultured HSCs was incubated with 100 ng/mL of leptin.The levels of SREBP-1c protein and mRNA were examined by Western Blot and real-time PCR,respesctively.The SREBP-1c promoter activity was analysed by transfection assay.Results: The results of the present study indicated that leptin significantly down-regulated the levels of SREBP-1c protein and mRNA.Furthermore,leptin also inhibited SREBP-1c promoter activity in HSCs.Conclusion: These results might provide new insights into the mechanisms of the unique role of leptin in the development of liver fibrosis.
