摘要
目的:建立柱前衍生化HPLC法测定左卡尼汀中光学异构体的含量。方法:左卡尼汀样品通过与衍生化试剂发生衍生化反应,用C18柱,以三乙胺缓冲溶液和四氢呋喃作为流动相,进行梯度洗脱,将左卡尼汀与其光学异构体分离;采用荧光检测器,激发波长234 nm,发射波长360 nm。结果:右卡尼汀在0.3~1.6μg.mL-1范围内线性关系良好,线性方程为Y=0.0173X+0.073 9r,=0.999 1,平均回收率为100.6%,RSD为1.97%(n=9)。结论:本法可用于检测左卡尼汀中光学异构体的含量,方法简便、准确、重现性好。
Objective: To develop a Pre-column derivatization HPLC method for the determination of D-carnitine in L-carnitine. Methods: Samples of L-carnitine were derivatized with ( + ) MNPC and analyzed on Cls column by fluorescence detector (excitation wavelength 234 nm, emission wavelength 360 nm) using triethylamine buffer and tetrahydrofuran as the mobile phase with gradient elution. Results: The linear range of D-carnitine is 0. 3 - 1.6μg·mL-1 ( Y= 0. 017 3X +0. 073 9, r =0. 999 1 ). The average recovery was 100. 6% with RSD 1.97% (n = 9). Conclusion: The method is appropriate for the determination of D-earnitine in L-earnitine. The method is simple, accurate and reproducible.
