摘要 目的研究DNA修复基因XRCC3多态性及其与结肠癌易感性的关系。方法选择80例经病理证实的结肠癌患者为肿瘤组,80例同期经门诊肠镜检查无异常的健康体检者为对照组,采用限制性片段长度多态性-聚合酶链反应(RFLP-PCR)法检测XRCC3基因C18607T的多态性分布,比较不同基因型与结肠癌易感性的关系。结果 XRCC3C18607T基因型C/C(野生型纯合子)、C/T(杂合子)和T/T(突变型纯合子)在肿瘤组的分布频率为58.75%、38.75%、2.50%,在对照组的分布频率为76.25%、22.50%、1.25%。各基因型在肿瘤组和对照组中的分布频率符合Hardy-Wein-berg平衡规律。与携带C/C的个体相比,携带T等位基因的变异基因型(C/T+T/T)的个体具有更高的结肠癌易感性。结论 XRCC3基因C18607T的多态性可能与结肠癌易感性相关。 Objective To study the relationship between the polymorphisms of DNA repair gene XRCC3 and susceptibility of colon carcinoma.Methods Selected eighty colon carcinoma patients as tumor cases and eighty health people as control cases.The XRCC3 C18607T genotypes were detected by restriction fragment length polymorphism-polymerase chain reaction(RFLP-PCR),and explored the relationship between the different genotypes and the susceptibility of colon carcinoma.Results The frequency of the XRCC3 C18607T genotypes C/C,C/T and T/T was 58.75%,38.75% and 2.50% respectively in colon carcinoma patients,but the frequency was 76.25%,22.50% and 1.25% respectively in normal control.The distribution of genotypes in colon carcinoma patients and the health controls was up to Hardy-Weinberg law.Compared with C/C genotype,combination of C/T and T/T genotype both increased the risk of colon cancer.Conclusion The polymorphism of XRCC3 C18607T may be associated with the increased risk of colon carcinoma.
