摘要 目的研究非生物、生物诱导子及两种诱导子协同作用对半夏细胞生长和总生物碱的积累。方法采用酸性染料比色法测定总生物碱的含量。结果添加1 mg/L Ag+、1 mg/L Cu2+和5 mg/L苯丙氨酸(Phe)总生物碱分别为:409,798和519μg/g,是对照的2.1倍,3.4倍和2.6倍;1 mg/L Ag+和10 mg/L Phe,1 mg/L Cu2+和5 mg/L Phe总生物碱含量是634和461μg/g,是对照的2.8倍和1.9倍;1 mg/L Cu2+诱导的愈伤组织半夏总生物碱含量最高。结论实验中的诱导子显著提高半夏总生物碱含量且不影响细胞生长。 Objective To study the influence of abiotic elicitors,bio-elicitors and their synergy on cell growth and total alkaloid accumulation in callus culture of Pinellia ternata.Methods The total alkaloids was tested by acid dye colorimetry.Results Total alkaloids were 409,798 and 519 μg/g when 1 mg/L Ag+,1 mg/L Cu2+ or 5 mg/L Phe were added in callus culture,which were 2.1、3.4 and 2.6 times of the control;total alkaloids were 634 μg/g and 461 μg/g by adding Two-factor inducer(1 mg/L Ag+ and 10 mg/L Phe,1 mg/L Cu2+ and 5 mg/L Phe),which were 2.8 and 1.9-fold of the control.The highest of total alkaloids was achieved by Cu2+ at 1 mg/L.Conclusion Elicitor can increase the total alkaloid content significantly and has little effect on cell growth.
