白花蛇舌草-半枝莲药对提取物抑菌活性部位研究 被引量:13

摘要 目的通过测定白花蛇舌草-半枝莲药对醋酸乙酯/95%乙醇(1∶1)提取上清液F0、析出浸膏F1、析出浸膏经AB-8大孔树脂富集后组分F2及F2经聚酰胺分离得到的各组分的抑菌活性,寻找药对提取物抑菌活性的活性部位或活性成分。方法采用一剂量法和二倍稀释法测定各提取对5种微生物(大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌、毛霉、酵母菌)的抑菌活性。结果与结论 F0抑制金黄色葡萄球菌活性最强;F1没有抑菌活性;F1经过大孔树脂富集后组分F2抑菌活性得到显著增强,其抑制枯草芽孢杆菌活性强于F0,抑菌效价值达8.37μg.ml-1;F2经聚酰胺分离得到的F2-10组分是抑制枯草芽孢杆菌的活性部位;各提取物对霉菌和酵母菌均无抑制活性。 Objective To investigate the antimicrobial activities of constituents from herb pair of Hedyotis diffusa Willd and Scutellaria barbata D.Don,including supernatant(F0) of their pair extract by ethyl acetate/95% ethanol(1:1),precipitation extract(F1),its enrichment(F2) by using AB-8 macroporous resin and all fractions separated from F2 by polyamide resin.Methods The standard curve assay was used to determine the antimicrobial activities of these extracts,regarding the biological activity as evaluation index.The tested species were Escherichia coli,Staphylococcus aureus,Bacillus subtilis,Mucro.sp.,Saccharom ycessp.Results and conclusion The supernatant of their pair extract showed the maximum activity to Staphylococcus aureus;the precipitation extract had no activity to tested species,but its enrichment was superior to supernatant in inhibiting Bacillus subtilis and its potency was 8.37 μg·ml-1.Fraction F2-10 which was separated from F2 was antimicrobial constituents of inhibiting Bacillus subtilis.All extracts had no activity to Mucro.sp.and Saccharom ycessp.
