摘要
目的研究黄粉虫蛋白提取物的纤溶活性及其性质。方法通过生物化学方法从黄粉虫Tenebrio molitorLinne组织分离纯化出纤溶活性蛋白,纤维蛋白平板法测定其纤溶性,对其进行溶血性、体外溶栓实验和血管生成抑制作用实验,并对其部分性质做了研究。结果分离纯化得到纤溶性蛋白。结论黄粉虫蛋白提取物具有纤溶性,该成分在40℃下基本稳定,最适反应温度为25~45℃,最适pH为7.5~8.0。其溶血率小于5%,不引起体外红细胞的明显溶血,安全性好。黄粉虫蛋白提取物受尿素和巯基乙醇(BME)的影响,而Na+,Mg2+,K+及Ca2+等各种金属离子及EDTA对其影响不大。
Objective To study the fibrinolytic activity of the active protein from Tenibrio molitor.Methods A novel of fibrinolytic protein from Tenibrio molitor was separated and purified by biochemical method,its effect on fibrin plate,fibrinolytic,hemocytolysis,angiogenesis and some properties were studied.Results The protein showed fibrinolytic activity.Conclusion The hemolysis rate of the protein from Tenibrio molitor is lower than 5%,so hemolytic crisis would not occur.The activity of the protein can be inhibited by urea and mercaptoethanol(BME),but can not be inhibited by Na+,Mg2+,K+ and Ca2+ and other metal ions and EDTA.The protein is stable under 40℃,its optimal temperature is 25~45 ℃,and the optimal pH is 7.5~8.0.
