摘要
目的探讨丹参注射液对血管内皮细胞低氧损伤的保护作用及可能的机制。方法将体外培养的人脐静脉内皮细胞株(Human Umbilical Vein Endothelial Cells,HUVEC)分为3组,即常氧培养组、低氧培养组及低氧培养+丹参组,通过倒置相差显微镜观察并比较各组细胞的形态变化;MTT法检测细胞活性;AO/EB染色和AnnexinⅤ-FITC/PI流式细胞术检测细胞凋亡;利用DCFH-DA荧光探针检测细胞内活性氧水平;并取各组细胞上清液进行总抗氧化能力检测。结果丹参能够显著减少低氧引起的细胞凋亡,提高细胞存活率,降低细胞内活性氧水平,并调节细胞总抗氧化能力。结论低氧条件下,丹参对HUVEC有保护作用,其机制可能与清除自由基有关。
Objective To study the protective effect of Danshen injection against hopoxia injury in human umbilical vein endothelial cells and possible mechanism.Methods Cultured human umbilical vein endothelial cells were divided into three groups,normal group,hypoxia group and hypoxia + Danshen group.The morphological changes of cells in each group were observed and compared with inverted phase contrast microscope.Viability of HUVEC was detected by MTT assay.AO / EB staining and Annexin Ⅴ-FITC/PI flow cytometry were used to analyze the apoptosis rate.DCFH-DA fluorescent probe was used to evaluate the intracellular reactive oxygen species level,and the total antioxidant capacity was also tested by ABTS assay.Results Danshen injection could ameliorate the hypoxia,induced apoptosis,increase cell viability,reduce the level of intracellular reactive oxygen species and regulate cellular total antioxidant capacity significantly.Conclusion Danshen injection has protective effects on HUVEC under hypoxis conditions,and its mechanism may be related to scavenging free radicals.
