没食子中没食子酸含量测定及总鞣质提取方法的研究 被引量:12

摘要 目的研究没食子总鞣质提取方法,测定没食子中没食子酸的含量。方法以提取物中总鞣质含量为指标,采用水浸泡、70%甲醇超声、50%乙醇搅拌、80%丙酮浸泡等提取方法提取得浸膏,用HPLC测没食子酸含量,用干酪素法测总鞣质含量。结果不同溶剂提取工艺中,鞣质提取率高低顺序为甲醇超声>水浸>乙醇搅拌>丙酮浸泡;没食子中没食子酸含量为2.25%。结论甲醇超声提取物中总鞣质含量最高,该方法可为提取没食子中总鞣质的提取方法。 Objective To investigate the extraction method and determine total tannins and gallic acids in Galla Turcica.Methods With the content of total tannins as index,tannin were extracted from Galla Turcica with aqueous methyl alcohol(70%,v/v) supersonic,water soakage,aqueous ethanol(50%,v/v) stir,and aqueous acetone(80%,v/v) soakage.Tannin was determined by ultraviolet spectrophotometer and gallic acid in Galla Turcica was determined by high performance liquid chromatography.Results In different solvent extraction methods,the extraction efficiency order was methyl alcohol supersonic,water soakage,ethanol stir,acetone soakage according to the content of tannin,the content of gallic acid in Galla Turcica was 2.25%.Conclusion The content of total tannins by alcohol supersonic is the highest compared with other extraction methods,and this method can be used in extraction of total tannins in the root of Galla Turcica.
