活血散结合剂中黄芪甲苷含量的测定

Content Determination of Astragalin Ⅳ in Huoxue Sanjie Granule by HPLC-ELSD

目的建立HPLC-ELSD法测定活血散结合剂中黄芪甲苷含量的方法。方法用shim-pack C18-ODS(150 mm×4.6mm,5μm)色谱柱,以乙腈-水(32:68)为流动相,流速为1.0 ml·min-1,柱温为30℃,检测温度为40℃,载气流量为4.06 L·min-1。结果黄芪甲苷进样量的线性范围是1.006～10.06μg,R=0.999 3,n=6;样品加样回收率为97.29%,RSD为1.37%(n=6)。结论该方法操作简便,重复性好,灵敏度高,专属性强。

Objective To eastablish an HPLC-ELSD method for the determination of Astragalin Ⅳ content in Huoxue Sanjie Granule. Methods The separation was performed on a shim-pack C18-ODS（150 mm×4.6mm,5μm） with acetonitrile-water（32：68） as mobile phase.The flow rate was set at 1.0 ml·min-1.Column temperature was 30℃,heater was 40℃.The air flow rate was 4.06 L·min-1. Results The linear range for astragalin Ⅳ was 1.006～10.06 μg,R=0.999 3,n=6.The recovery ratio of astragalin Ⅳ was 97.29% and RSD was 1.37%（n=9）. Conclusion The method is simple,rapid,reprodu cible,sensitive,accurate and is suitable for the determination of astragalin Ⅳ.
