摘要
目的观察姜黄素对单侧输尿管梗阻(UUO)大鼠肾组织细胞外基质成分胶原蛋白-Ⅰ(Collagen-Ⅰ,Col-Ⅰ)、胶原蛋白-Ⅲ(Collagen-Ⅲ,Col-Ⅲ)和纤维连接蛋白(Fibronectin,FN)表达的影响,分析其抗纤维化的机制。方法采用大鼠单侧输尿管梗阻(UUO)模型。将32只SD大鼠随机分为假手术组,模型组,姜黄素1组和姜黄素2组。对模型组和治疗组大鼠行左侧输尿管结扎建立单侧输尿管梗阻的动物模型。姜黄素1治疗组于术后第2天给予姜黄素100mg.kg-1.d-1灌胃,姜黄素2治疗组于术后第10天给予姜黄素100 mg.kg-1.d-1灌胃。术后28 d处死大鼠,取梗阻侧肾组织行HE和Masson染色以观察各组大鼠肾组织病理改变。用免疫组化染色方法观察Col-Ⅰ,Col-Ⅲ和FN的表达分布。结果模型组肾小管间质损伤程度、胶原的表达均高于假手术组。姜黄素能明显改善大鼠肾脏病理改变,减少细胞外基质沉积。结论姜黄素可通过下调Col-Ⅰ,Col-Ⅲ和FN在肾组织的表达,从而减少细胞外基质的过度聚集,抑制或减缓肾小管间质纤维化的发生发展。
ObjectiveTo study the effect of curcumin on the expression of Col-Ⅰ,Col-Ⅲ and FN,in umilateral uteral obstrution rats.Methods32 healthy Sprague-Dawley rats were randomly divided into 4 groups: sham-operation group,UUO group,curcumin group 1 and curcumin group 2.UUO group rats were treated with left unilateral uteral obstruction.2 days after surgery,the rats in curcumin group 1 were intragastrically administered with curcumin(100 mg·kg-1·d-1);10 days after surgery,the rats in curcumin group 2 were intragastrically administered with curcumin(100 mg·kg-1·d-1).The sham-operation group and UUO group were intraperitoneally injected with identical volume of normal saline.28 days after surgery,obstructed-kidneys were harvested.The dynamic histological changes of renal interstitial tissues were observed by HE and Masson staining.Col-Ⅰ,Col-Ⅲ and FN were observed by immunohistochemical method.ResultsThe tubulointerstitium damage degree and collagen expression in UUO group were increased compared with sham-operation group.However,these morphological changes and excessive extracellular matrix were notably reduced after treatment with curcumin.ConclusionCurcumin can reduce the excessive extracellular matrix,inhibit the development of tubulointerstitium fibrosis by decreasing the expression of Col-Ⅰ,Col-Ⅲ and FN.
