姜黄素对大鼠脊髓缺血再灌注损伤后NF-κB和Hsp70的表达研究 被引量:10

摘要 目的研究姜黄素对大鼠脊髓缺血再灌注损伤后NF-κB和Hsp70的表达,进一步明确其作用机制。方法 72只雄性SD大鼠随机分为姜黄素实验组、缺血模型组和正常对照组,每组24只,前两组经夹闭左右肾动脉之间腹主动脉制作脊髓缺血再灌注损伤模型,对照组不做任何处理。实验组缺血前10 min舌下静脉注射20mg/kg姜黄素;模型组缺血前10min舌下静脉注射同等量生理盐水;对照组经舌下静脉注射等量的生理盐水。再灌注3,12,24,72h不同时间点各取6只大鼠腰段脊髓组织,分别经HE染色、免疫组织化学法和Western blot法检测NF-κB和Hsp70的表达。结果 NF-κB、Hsp70在前两组脊髓组织中的表达均有统计学意义(P<0.05),姜黄素对各时间点脊髓组织中NF-κB表达的抑制率分别是13.12%,15.14%,19.18%和18.02%;同时可诱导Hsp70蛋白的表达,对Hsp70的诱导率分别为10.40%,12.71%,15.95%和13.18%。结论姜黄素对大鼠脊髓缺血再灌注损伤后可抑制早期炎症反应的发生,同时对脊髓缺血再灌注损伤具有保护作用,调控Hsp70的表达可能是其作用机制之一。
