摘要
目的研究外感温燥对BALB/c小鼠气道muc5ac表达的影响,探讨温燥影响气道粘液分泌的规律。方法 SPF级BALB/c小鼠80只,雌雄各半,随机分为4组:正常对照组(C组)、温燥模型Ⅰ组(WⅠ组)、温燥模型Ⅱ组(WⅡ组)、桑杏汤治疗组(S组),置人工气候箱内按"温度-相对湿度-风"条件综合刺激进行造模。采用ELISA法检测支气管肺泡灌洗液(BALF)中TNF-α的蛋白表达量,分别用实时定量荧光PCR法和免疫组化法分别检测muc5ac mRNA和蛋白在小鼠气道的表达。结果 WⅠ组小鼠BALF中TNF-α含量和气道muc5ac mRNA及其蛋白表达明显高于C组(P(0.01),WⅡ组比WⅠ组显著增多(P(0.01),S组明显少于WⅠ组(P(0.01)。Spearman等级相关分析结果显示,小鼠BALF中TNF-α蛋白含量与气道muc5ac mRNA、及蛋白的表达均呈正相关(r=0.86,P(0.01;r=0.91,P(0.01)。结论温燥可增加BALB/c小鼠气道muc5ac mRNA及其蛋白的表达,TNF-α可能参与了该过程的调节。
ObjectiveTo study the effect of warm-dryness on muc5ac expression of airway and law of airway mucus secretion in BALB/c mice.Methods80 specific pathogen Free BALB/c mice,male and female in half,were randomly divided into 4 groups: normal control group(C group),warm-dryness modelⅠ(WⅠgroup),warm-dryness model Ⅱ(W Ⅱ group) and SangXingTang treatment group(S group),and the model was induced in "temperature-relative humidity-the wind" conditions.TNF-α protein expression in bronchoalveolar lavage fluid(BALF) was detected by ELISA,and expression of muc5ac mRNA and muc5ac apomucin in airway was detected by real time quantitative fluorescence PCR and immunohistochemical assay,respectively.ResultsTNF-α level and expression of muc5ac mRNA and muc5ac apomucin in WⅠgroup were higher than in C group(P〈0.01),W Ⅱ group was higher than WⅠgroup(P〈0.01),and S group than those in WⅠgroup(P〈0.01).Conclusion Warm-dryness can increase the expression of muc5ac mRNA and muc5ac apomucin,and TNF-α might be involved in the regulation of this process.
