摘要
目的:优化用硅胶柱层析提取连翘叶中连翘酯苷的最佳提取工艺。方法:采用L9(34)正交试验设计,考察上样量(A)、洗脱液流速(B)和柱床径高比(C)对连翘酯苷得率的影响,优选最佳提取工艺条件。结果:上样量、洗脱液流速和柱床径高比对连翘酯苷得率的影响大小依次为:C>A>B,最佳工艺为样品与硅胶用量比1∶20,洗脱液流速1.5BV/h,柱床径高比1∶10。结论:该制备工艺简便、稳定和可行。
Objective:To optimize the extraction process of forsythiaside by silica gel column chromatography.Methods:Orthogonal design [L9(34)] was used to select the best extraction process.The loading quantity of sample(A),eluent flow rate(B) and diameter-height ratio of column(C) were set as indexes for orthogonal test.Results: Diameter-height ratio of column(C) was the first influential factor,the loading quantity(A) of sample was the second and the eluent flow rate(B) was the third.The optimal extraction process was achieved by loading quantity of sample 1∶20,the eluent flow rate 1.5 BV/h and diameter-height ratio of column 1∶10.Conclusion: The optimal extraction process of forsythiaside is simple,stable and feasible.
