菟丝子含药血清对成骨细胞代谢调控的影响 被引量:11

Effect of Dodder-Containing Serum on the Metabolism of Osteoblasts In Vitro

摘要 目的探讨菟丝子含药血清对体外培养大鼠成骨细胞代谢调控的影响。方法采用血清药理学方法制备菟丝子含药血清;采用改良的组织块法体外培养大鼠成骨细胞;分别用5%、10%、15%、20%菟丝子含药血清诱导培养成骨细胞24h、48h、72h后,MTT法检测大鼠成骨细胞的增殖情况;用对硝基苯二钠基质动力学法(PNPP)测定细胞内碱性磷酸酶(ALP)的活性;培养48h后用RT-PCR方法检测Ⅰ型胶原(COL-Ⅰ)mRNA的表达。结果 5%和10%菟丝子含药血清在48h和72h能显著促进成骨细胞增殖并提高ALP活性;在48h时10%菟丝子含药血清可促进COL-ⅠmRNA的表达。结论 10%菟丝子含药血清能够明显促进大鼠成骨细胞的增殖与分化,表明其具有促进成骨作用。 Objective To explore the effect of Dodder-containing serum on the metabolism of rat osteoblasts(OB) cultured in vitro.Methods The Dodder-containing serum was prepared by serological pharmacological method,and the improved tissue block culture was adopted to culture the osteoblasts in vitro of newly born SD rats.After the culture of OB(n=8) for 24,48,and 72 hours with 5%,10%,15%,and 20% Dodder-containing serum respectively,the MTT method was used to observe the proliferation,the PNPP matrical dynamic method was applied for observing the activity of intracellular alkaline phosphatase(ALP),and after 48h culture,the RT-PCR technique was employed to detect the COL-I mRNA expression.Results After 48h and 72h culture,the 5% and 10% Dodder-containing serum functioned significantly in improving the proliferation of OB and enhancing the activity of ALP.After 48h culture,the 10% Dodder-containing serum functioned in promoting the COL-I mRNA expression.Conclusion The 10% Dodder-containing serum can significantly promote the proliferation and differentiation of OB,indicating that it benefits osteogenesis.
