茯苓菌丝体多糖径向流色谱分离参数优化 被引量:4

摘要 在单因素试验的基础上,通过Box-Benhnken组合设计和响应曲面分析法优化茯苓菌丝体多糖径向流色谱分离的工艺条件。响应面分析结果表明,当样品浓度为8 mg/mL,上样量为40 mL,上样流速为25 mL/min,洗脱流速为33 mL/min时,多糖回收率达到83.26%,比传统轴向色谱分离及化学方法效率更高,时间更短。 Seperation technology was tested by radial flow chromatography (RFC) packed with ion-exchange. Based on the single factor test, response surface method (RSM) was employed to obtain the best result. The purification efficiency of RFC was compared with that of axial column and traditional chemical method. RSM analysis showed that the optimal conditions for the purification of Poria cocos polysaccharides with RFC were as follows: sample concentration of 8 mg/mL, sample volume of 40 mL, sample flow-rate of 25 ml./min and elution flow-rate of 33 ml/min. The comparative test results indicated that the best purification efficiency was obtained by RFC within the shortest time.
